Copper is an essential co-factor for all organisms , but when its concentration exceeds a certain threshold, it will have a toxic effect and further induces cell death. Todd R. Golub’s group of the Broad Institute, in collaboration with Peter Tsvetkov (first author), published a paper to come up with a new mode of cell death which is copper-depended and regulated on mitochondrial respiration. Copper directly binding with lipoylated components of TCA cycle leads to abnormal aggregation of fatty acylated proteins and loss of iron-sulfur clusterin, resulting in proteotoxic stress responses that ultimately lead to cell death.
